Extracellular vesicles derived miRNAs from bovine milk modulate the inflammatory response to lipopolysaccharide in vitro

چکیده

Abstract Bovine milk is composed of a variety biologically active compounds including mi-croRNAs (miRNA), which are found within extracellular vesicles (EVs). Extracellular vesicle-derived miRNAs may play crucial role in the regulation immune response. Thus, this study aimed to identify specific present bovine that modulate system’s response infection. To address this, RAW-Blue™ Cells, mouse macrophage reporter cell line was challenged with E. coli lipopolysaccharide (LPS) and incubated pool isolated from through 24-hour period. After incubation, QUANTI-Blue™ assay used detect activation transcription factor NF-κB by different treatments. Raw-Blue™ cells treated induced some extend signaling pathways leading relative control (P<0.01). Bovine-milk derived suppressed LPS-induced activity dose-dependent manner (P=0.01). top expressed driving such effects vitro, real-time qPCR assays were performed samples. The miRNA-profiling revealed presence involved tumor suppression apoptosis. Overall, our preliminary findings indicated have potential LPS vitro. Texas Tech University, School Veterinary Medicine

Biocompatibility of highly purified bovine milk-derived extracellular vesicles

Extracellular vesicles (EVs) deliver biologically active cargos from donor cells to recipient cells for intercellular communication. Since the existence of RNA cargo was discovered, EVs have been considered to be useful drug-delivery systems. Specifically, EVs from bovine milk (mEV) are one of the most promising platforms, since bovine milk is a scalable source of EVs for mass production. Howev...

Extracellular Vesicles Derived from Gastrointestinal Microbiota: A New Approach to Clinical Studies

Extracellular vesicles, naturally released from all cell types including bacteria, are of great importance in medical microbiology due to transporting a variety of biomaterials, enzymes, and virulence factors, regulating immunity, and having roles in colonization and initiation of signaling pathways. These vesicles are also secreted from microbiota in the gastrointestinal tract and affect the h...

Extracellular Vesicles Derived from Acholeplasma laidlawii PG8

Extracellular vesicle production is believed to be a ubiquitous process in bacteria, but the data on such a process in Mollicutes are absent. We report the isolation of ultramicroforms - extracellular vesicles from supernatants of Acholeplasma laidlawii PG8 (ubiquitous mycoplasma; the main contaminant of cell culture). Considering sizes, morphology, and ultrastructural organization, the ultrami...
